我院醫(yī)學(xué)檢驗(yàn)系潘曉玲教授、李曉丹副教授、葉湘漓教授等擬基于既往研究成果申報(bào)2024年湖南省預(yù)防醫(yī)學(xué)科學(xué)技術(shù)獎(jiǎng),現(xiàn)公示如下:
項(xiàng)目名稱:蚊媒病毒新型防控技術(shù)和研究平臺(tái)。

項(xiàng)目簡(jiǎn)介:蚊媒病毒是一類由蚊蟲叮咬傳播的病毒,包含多種引起人類病毒性腦炎或出血性疾病的病原體。本項(xiàng)目從蚊媒和病毒兩方面探索蚊媒病毒的新型媒介防控策略和高效抗病毒研究平臺(tái),為蚊媒病毒的控制和治療提供科技支撐。蚊媒方面,本項(xiàng)目系統(tǒng)研究了一種可導(dǎo)致蚊蟲不育及抗病毒效應(yīng)的昆蟲共生菌——沃爾巴克氏體(Wolbachia)的抗病毒機(jī)制,發(fā)現(xiàn)其可通過調(diào)節(jié)埃及伊蚊(登革病毒、寨卡病毒、基孔肯雅病毒傳播媒介)細(xì)胞長鏈非編碼RNA(lncRNAs)激活蚊媒體內(nèi)先天免疫通路,促進(jìn)其與宿主的共生關(guān)系,并賦予宿主抗感染的作用?;?em>Wolbachia致蚊蟲不育和抗病毒作用,本項(xiàng)目首次在長沙市芙蓉區(qū)釋放人工感染Wolbachia的白紋伊蚊雄蚊,顯著降低了投放區(qū)域內(nèi)白紋伊蚊(登革病毒、基孔肯雅病毒傳播媒介)的數(shù)量,蚊子叮咬率也大幅降低,對(duì)預(yù)防蚊媒病毒感染有重要意義。病毒方面,本項(xiàng)目以重要的蚊媒病毒反向遺傳學(xué)平臺(tái)為基礎(chǔ),開展病毒復(fù)制機(jī)制及抗病毒研究:首次發(fā)現(xiàn)寨卡病毒RNA環(huán)化元件“DAR”對(duì)病毒復(fù)制的影響具有種屬特異性,與病毒的跨種傳播相關(guān);成功構(gòu)建了一種新發(fā)蚊媒病毒馬亞羅病毒的報(bào)告病毒系統(tǒng),并以此為工具建立了便捷高效的抗病毒藥物篩選平臺(tái)。
